BNF | Development of screening assays for immunomodulatory proteins
Switchkine is a start-up developing cytokine-based therapies to restore the balance in chronic inflammatory disease. Many cytokines exhibit reduced efficacy at low pH, an environment typical of inflammatory sites. At Switchkine we are engineering pH-switchable cytokines to enhance local activity at the target site.
The goal of this project is to setup and validate assays using primary human immune cells such as monocytes, macrophages, DCs and lymphocytes, to screen existing and future cytokine constructs developed by Switchkine. In collaboration with the team, you will develop assays to analize the changes in phenotype and functionality of different cell types in response to treatment with our cytokines. You will be part of the process of research in a start-up environment, from experimental design to data integration and presentation.
The main tasks will be the following:
- Processing of human buffycoats
- Culture and differentiation of different human immune cell types
- Perform in vitro assays to analyze cell phenotype and funtionality by flow cytometry and immunoassays
- Participate to experimental design
- Data analysis, interpretation and presentation
If you join us for this project, you will learn several aspects of the development of molecules and their validation for use as drug candidates. Such processes are fundamental for both fundraising and application to regulatory authorities for the initiation of clinical studies. Switchkine being a small start-up, it will allow you to experience how the different functions interact and have a deeper understanding of this environment.
Requirements:
- Understanding of human immunology, macrophages and T cells biology
- Isolation and culture of human immune cells
- Conventional multicolor flow cytometry
- Immunoassays (ELISA, ELIspots, Luminex…)
- Experience in data analysis (FlowJo, Prism...)
- Co-culture of immune cells or in vitro model of inflammation is a plus
- Signaling pathway analysis is a plus
- English spoken and written
Requested trainning:
Master or PhD in Life sciences with specialization in Immunology
